Plasma sampling Sample Clauses

Plasma sampling. 100 µL facial vein blood sampling before Tamoxifen induction and every 30 days will be processed to plasma for both treated and untreated mice. The PI will provide part of these samples to Minoryx to store or carry out the analysis of circulating MIN-102 amount and adiponectin. After euthanasia or sacrifice, a terminal plasma sample will be collected. Brain will be harvested and divided longitudinally: one half will be flash frozen in liquid nitrogen for molecular and biochemical investigations, the other half will be fixed in PFA 4%, cryopreserved in 30% sucrose and frozen in isopentane for histological evaluations. Experiment 2: in vivo evaluation of MIN-102 treatment in pre-symptomatic mice for histology/molecular biology Treatment: starting from the first day after Tamoxifen induction, 12 SLICK-Coasy (3 males and 3 females, treated and not treated), and 4 CTRL mice (2 males and 2 females, not treated) will be administered, when applicable, with a MIN-102 supplemented diet at a dose of 75 mg/kg/day. This regimen will be followed up as long as mice are in good clinical conditions as concerning weight gain, motor performance and overall health. All animals will be sacrificed at the same time point (2 mpi). Experiment 3: ex vivo evaluation of MIN-102 treatment efficacy Samples collected during Experiment 1 and 2 will be used to investigate the effect of MIN-102 on molecular, biochemical and histological signs related to the disease.
