Digestion. Restriction endonucleases were used to digest PCR products for genotyping or SSCP. PCR products were estimated to contain DNA at a concentration of 12 ng/ul. HhaI: PCR product 8ul 10 x buffer C 1ul HhaI 1ul 37°C overnight. Deactivation, 95°5 minutes. BsrI: PCR product 21.5ul 10 x buffer IV 2.5ul BsrI 1ul 65°C 2 hours. Not heat deactivated. PstI/PvuII: PCR product 13ul 10 x buffer 3 1.5ul PstI 0.25ul (20 units) PvuII 0.25ul (40 units) 37°C overnight. Not heat deactivated.
Digestion. Blended primary sludge and WAS is sent to seven anaerobic digestion tanks with a total capacity of 9.8 million gallons. The digestion tanks include floating covers for gas storage. Gas produced from digestion is sent to a cogeneration facility for energy and heat production.
Digestion. This task will review historical solids loading rates, biogas production, and other operating parameters as available and provided by the City. The operation of the anaerobic digestion system will be evaluated with respect to opportunities for improving volatile solids reduction and biogas production. It is assumed that opportunities for improvements will be limited to the heat recovery and mixing systems. An overall condition assessment of the tanks, covers, and ancillary components will not be performed.
