Mice. Female C57BL/6 (H-2b) mice were obtained from Xxxxxxx River Labora- tories (L’Arbresle, France) and kept under standardized conditions in the Leiden University Medical Centre animal facility. The in vivo experiment as described in section 2.8 was carried out according to the Dutch Experiments on Animal Act, which serves the implementation of “Guidelines on the protection of experimen- tal animals” by the Council of Europe. 7

(a) MAYO will provide to LICENSEE three (3) SPF breeder male mice and, depending on availability, up to fifteen (15) male or female mice of strain Tg 2576 of the Stock within 60 days of the Effective Date of this LSRA. In the event that interest, in the form of signed license and sponsored research agreements (such as this LSRA) received by MAYO, exceeds MAYO’s ability to deliver Stock, then such Stock will be provided on a first-come basis based on the date of receipt of the signed license and sponsored research agreements. In the event the projected delay shall exceed 30 days beyond the 60-day estimated period, LICENSEE shall have the right to terminate this LSRA.

Mice. Age- and sex-matched C57BL/6 (B6) mice were purchased from The Xxxxxxx Laboratory (JAX) to specifically control the SFB status of recipient mice in our SFB-containing microbiota transfer studies. All SFB-containing microbiota transfer studies were initiated within 2 weeks of arrival from JAX in order to avoid unintentional colonization by SFB in our animal facility. Importantly, immediately prior to the introduction of SFB-containing microbiota, mice were verified to be void of SFB (as determined by qPCR detection for SFB DNA in cecal contents and fecal pellets). B6 mice purchased from Taconic were utilized as donors of homogenized cecal contents for SFB-containing microbiota transfer experiments. Mice were maintained under specific pathogen-free conditions and animal protocols were reviewed and approved by the Institute Animal Care and Use Committee of Georgia State University. Antibodies and reagents The following antibodies were purchased from eBioscience: IFNγ (XMG1.2), IL- 17A (eBio17B7), CD25 (PC61.5), CD3ε (eBio500A2), and RORγ(t)-PE (B2D). Antibodies purchased from BD Biosciences were: TCRβ (H57-597), IL-17A (TC11-18H10), and CD4 (RM4-5). Dead cells were identified using the fixable Aqua dead cell staining kit (Invitrogen). Isolation of LP cells and flow cytometry was performed as previously described (126). Neutralizing antibodies for anti- mouse IL-1β (B122), anti-mouse IL-1R1 (BE0256) and isotype control (Hamster IgG) were purchased from BioXCell. Anti-mouse Siglec-F (238047) and isotype control (rat IgG) were purchased from R&D SYSTEMS. Specific microbiota transfer studies The cecal contents from Taconic B6 mice were resuspended in 5 ml of sterile PBS, filtered through a 100 µm cell strainer, and 150 ul of the homogenate was gavaged twice into each mouse with 3 hr between each gavage. The homogenate was verified to contain SFB via qPCR (29). Recipient mice were utilized within two weeks of arrival from the JAX and verified to be void of SFB prior to gavage and colonized post-gavage based on qPCR analysis of fresh fecal pellets. Mice were euthanized and organs were harvested to assess Th17 cell differentiation at 1 week post-gavage. Air tickets and hotel rooms wholesale Safe2Travel’s air tickets and hotel rooms wholesale business represents the sale of air tickets and hotel rooms on behalf of suppliers to customers including other travel agents. To maintain steady supply of travel products, air tickets and hotel rooms are occasionally purchased from the suppliers and sold to retail customers during festive periods when travel demand is especially high. Safe2Travel serves as an efficient distribution channel for its partner arlines to maximise their overall revenues and utilisation of their aircrafts. Being the air ticket consolidators for these airlines, an incentive bonus is received if Safe2Travel achieves a certain sales volume as prescribed by these airlines. Corporate leisure The corporate leisure division offers a full range of products and services for the corporate travel customers’ vacation and personal trips such as air ticketing, hotel accommodation, land transportation and vacation packages.
