Transformation of E coli Chemically-competent DH5α cells (NEB) were thawed on ice (typically 25 μL per transformation). The plasmid DNA or ligation reaction to be transformed was then mixed with the cells and left on ice for 15 minutes. For ligation reactions, 5 μL of the reaction volume was used for the transformation; for plasmid DNA, approximately 50 ng of DNA. The cells were then incubated at 42°C for 45 seconds and then placed back on ice for 2-3 minutes. Following this, the cells were resuspended in 75 μL of SOC medium and incubated for an hour at 37°C. For transformations of ligation reactions, the entire transformation mixture was plated onto selective LB plates and grown overnight at 37°C. For plasmid transformations, 1/10 of the reaction mixture was plated.

  • Finalization of Evaluation A Written Report 1 Before the evaluation cycle is final, and not later than May 10, a copy of the formal written evaluation report shall be given to the teacher and a conference shall be held between the teacher and the evaluator.

Channelization can be accomplished through the use of a multiplexer or a digital cross-connect system at the discretion of BellSouth. Once UC has been installed, NewPhone may request channel activation on a channelized facility and BellSouth shall connect the requested facilities via COCIs. The COCI must be compatible with the lower capacity facility and ordered with the lower capacity facility. This service is available as defined in NECA 4.

  • Implementation of Strategic Plan Goals This Agreement supports the County’s Strategic Plan, Goal 1, Operational Effectiveness/Fiscal Sustainability. This Agreement will provide revenue reimbursement to the Department for services rendered.

  • Prevention of Avoidance The Parties shall implement as necessary requirements to prevent Financial Institutions from adopting practices intended to circumvent the reporting required under this Agreement.

  • Cessation of Business Any Obligor suspends or ceases to carry on (or threatens to suspend or cease to carry on) all or a material part of its business.
