Strand displacement by DNA polymerase Sample Clauses

Strand displacement by DNA polymerase δ In S. cerevisiae, DNA polymerase δ is a complex consisting of three subunits: Pol3, Pol31, and Pol32 (Xxxxx et al. 1998). Pol3 is the catalytic subunit and it contains both polymerase and exonuclease (3’-5’) activities (Xxxxxxx and Burgers, 2016). Furthermore, all three subunits contain PIP-box motifs (QxxhxxFF) for interacting with PCNA (Xx et al. 2002; Xxxxxxx et al. 2011). Interactions with PCNA have been shown to increase the processivity of both DNA polymerase δ and DNA polymerase ε in vitro (Xxxxxxxx et al. 2007; Xxxxxx et al. 2017). During replication on the lagging strand, PCNA is loaded onto primer termini by RFC in a reaction that requires the hydrolysis of ATP (Xxxxxxxxx and Xxxxxxxx, 1990). DNA polymerase δ interacts with PCNA to extend primers synthesised by DNA polymerase α/primase until it reaches the 5’-end of a downstream Okazaki fragment, at which point it begins to displace the RNA/DNA. Importantly, the rate of synthesis decreases with each displaced nucleotide. This ensures that the flaps generated by the polymerase do not become too long for downstream processing (Burgers and Xxxxxx, 2017). In addition, DNA polymerase δ has been shown to switch between its polymerase and nuclease activities, leading to a cycle of continuous elongation and degradation of the nascent DNA (Burgers, 2009). This process has been termed ‘idling’ and is another mechanism through which the polymerase is restricted from excessive strand displacement. Unlike DNA polymerase δ, DNA polymerase ε is very poor at strand displacement synthesis (Xxxxxxxxx et al. 2014; Xxxx et al. 2004; Xxxxxx et al. 1993). Recently, it was shown that DNA polymerase ε is unable to carry out extensive strand displacement synthesis unless its 3’-5’ exonuclease activity is first removed (Xxxxx et al. 2016).

  • Permitted Uses and Disclosures of Phi by Business Associate Except as otherwise indicated in this Agreement, Business Associate may use or disclose PHI, inclusive of de-identified data derived from such PHI, only to perform functions, activities or services specified in this Agreement on behalf of DHCS, provided that such use or disclosure would not violate HIPAA or other applicable laws if done by DHCS.

  • Loop Provisioning Involving Integrated Digital Loop Carriers 2.6.1 Where InterGlobe has requested an Unbundled Loop and BellSouth uses IDLC systems to provide the local service to the End User and BellSouth has a suitable alternate facility available, BellSouth will make such alternative facilities available to InterGlobe. If a suitable alternative facility is not available, then to the extent it is technically feasible, BellSouth will implement one of the following alternative arrangements for InterGlobe (e.g. hairpinning):

  • Effective Date Term Termination and Disconnection 3.1 Effective Date 3.2 Term of Agreement 3.3 Termination

  • PERMITTED USES AND DISCLOSURES BY CONTRACTOR Except as otherwise limited in this Schedule, Contractor may use or disclose Protected Health Information to perform functions, activities, or services for, or on behalf of, County as specified in the Agreement; provided that such use or disclosure would not violate the Privacy Rule if done by County.

  • Permitted Uses and Disclosures by Business Associate 1. Business Associate may only use or disclose protected health information as necessary to perform the services as outlined in the underlying agreement.

  • Technological Displacement The Employer agrees that, whenever possible, no employee shall lose employment because of technological change, utilizing normal turnover of staff to absorb such displaced employees. However, when necessary to reduce staff, it shall be done as outlined in Article 6.01.

  • Undue Burden and Fundamental Alteration For any technology-related requirement in this Agreement for which the Recipient asserts an undue burden or fundamental alteration defense, such assertion may only be made by the Superintendent or by an individual designated by the Superintendent and who has budgetary authority after considering all resources available for use in the funding and operation of the service, program, or activity, and must be accompanied by a written statement of the reasons for reaching that conclusion, including the cost of meeting the requirement and the available funding and other resources. The written statement will be certified by the determining official. If such a determination is made, the certifying official will describe in the written statement how it will provide equally effective alternate access, i.e., other action that would not result in such an alteration or such burdens but would nevertheless ensure that, to the maximum extent possible, individuals with disabilities receive the same benefits or services provided by the Recipient as their nondisabled peers.

  • Integrated Digital Loop Carriers The feeder portion of some loops may be provide by means of Integrated Digital Loop Carrier (IDLC). IDLC provides a fiber optic cable transmission path that travels directly into BellSouth’s central office local switch. Where BellSouth uses IDLC ,if technically feasible and capacity does exist, BST will provide Al-Call with a Designed DS0 UVL by using alternative provisioning techniques including but not limited to such as “hairpinning” and DAC grooming. Alternative provisioning techniques will be provided at no additional cost to Al-Call . Hairpinning involves providing a DS0 signal from an IDLC-served loop to Al-Call ’s collocation equipment by using a dedicated pathway that traverses BellSouth’s central office switch. BellSouth will provide such DS0 signal to Al-Call by establishing a copper cross connect between the BellSouth switch and Al-Call ’s collocation equipment.
